Mechanism of action of binary antisense oligonucleotides (biASOs)

In the presence of activator RNA, ASOa and ASOb strands assemble to form a four-way junction (4WJ) structure with the target mRNA of a housekeeping gene. This 4WJ complex becomes a substrate for RNase H, leading to target mRNA cleavage and release of biASO components for further catalytic activity. Adapted from [6]
